Pule Pandak is a traditional plantation that have a high economic and health values. It is categorized as an endangered species, but it seems that there are lack of efforts to save the species from extinction. From those alkaloids, Reserpine is the most important alkaloids in hypertension treatment. Pule pandak is an endangered species that lives in a forest, so action to save the species must be done immediately. These research is aim to find the scientific foundation about the compatibility of grafting between R. serpentine and R. verticilata with morphological and anatomical observations. This research was conducted Ngaglik sub-district, Sleman district by using following treatments: Shading spacing were 25%, 50%, and 75%. Grafting methods: 1). Approach side grafting method and 2). Side grafting method. Succesful grafting with scores 2 – 4 treatment with bokasi menure 200 g/plant, Shading spacing were 50%, and 75%.Limited in this study can be concluded that
1. Grafting between Ruvolfia verticilata and Rauvolfia serpentina and is compatible 2. Method of side grafting is better than approach side grafting 3. Shade level that is conducive to the formation of unions is 50% – 75% with humidity levels of 80% – 90% and air temperature 28 0C – 260C 4. Up to the age of 6 months after rootstock cutting, there are inhibition of growth anddried root weight, but the levels reserpina increased
